摘要 目的 探讨大蒜素对小鼠黑素瘤细胞B16-F1增殖及凋亡的影响。方法 传代培养小鼠黑素瘤B16-F1细胞,使细胞浓度为5×10^4个/ml,分别加入不同浓度(3,6,9,12,15μg/m1)的大蒜素细胞培养液。培养12,24,36,48h后,噻唑蓝(MTT)法检测细胞生存活性,原位末端标记(TUNEL)法检测细胞凋亡,定量RT—PCR法检测ras基因的表达水平。结果 3~15μg/ml的大蒜素对B16-F1细胞均有杀伤作用,当大蒜素的浓度为3~6μg/ml,诱导肿瘤细胞凋亡,且呈浓度时间相关性。当大蒜素的浓度较高时,细胞呈中毒反应,凋亡作用不明显。3~15μg/ml的大蒜素下调ras基因的表达,并且呈显著的浓度相关性。结论 大蒜素对小鼠黑素瘤B16-F1细胞有杀伤作用,可以明显诱导肿瘤细胞的凋亡。 Objective To study the effect of allicin on growth and apoptosis of mouse ascites melanoma cell line B16-F1. Methods After exposed to different concentration of allicin for different time,the survival rate of BI6-F1 eells was explored by MTT, Terminal deoxylnucleotide transferase directed X-UTP nick and end labelling (TUNEL) was used to observe apoptosis, RTPCR was used to study the expression of ras gene, Results Compared with that of the experimental control group,allicin showed significant inhibiting effect on the growth BI6-F1 cells and the expression of ras gene at the dosage of 3 - 15 μg/ml. From 3μg/ml to 6μg/ml concentration of allicin,the apoptotic rate of cells was significantly increased,the expression of ras gene was significantly decreased. Conclusion Allicin can inhibit the growth of mouse ascites melanoma cell line B16-F1 and induce the apoptosis. The expression of ras gene was decreased by allicin.
