胃癌组织和胃周淋巴结COX-2mRNA的表达及其意义 被引量:6

摘要 目的:探讨胃癌组织和胃周淋巴结内环氧化酶-2(COX-2)mRNA的表达特点及其意义。方法:选取胃癌患者43例和正常胃粘膜组织15例,取癌灶、癌旁、淋巴结和正常胃粘膜标本,通过采用PCR(Real-timeSemi-quantitativePCR)方法检测COX-2mRNA表达。结果:1)癌灶处COX-2mRNA表达高于癌旁和正常胃粘膜组织的表达;2)癌旁COX-2mRNA表达高于正常胃粘膜组织的表达(P<0.05);3)侵透浆膜的胃癌组织COX-2mRNA表达高于未透浆膜的表达;4)弥漫生长的胃癌组织COX-2mRNA表达高于团块状及巢状生长者的表达(P<0.05);5)淋巴结转移阳性的淋巴结组织COX-2表达高于淋巴结转移阴性的表达(P<0.05)。结论:胃癌组织COX-2mRNA的表达明显升高,其表达与胃癌生物学行为相关;胃癌周围淋巴结组织COX-2mRNA的表达情况可作为推测癌周淋巴结有无微小转移癌的一个具有提示意义的参考指标。 Objective: To explore the characteristics and significance of cyclooxygenase-2 (COX- 2)mRNA expressions in gastric cancer and lymph node (LN). Methods: 43 patients with gastric cancer and 15 normal controls were chosen. The specimens were obtained from gastric cancer, adjacent normal mucosa, LN and normal gastric mucosa biopsy (control group) by upper gastrointestinal endoscopy. mRNA was extracted from specimens to measure the COX-2 gene mRNA levels by real-time PCR method. Results: 1) Our results revealed that COX-2 levels in gastric cancer were significantly higher than those in adjacent normal mucosa and control group; 2) COX-2 levels were significantly higher in adjacent normal mucosa than in control group; 3) Gastric cancer COX-2 levels were significantly higher in infiltrated serosa type than in uninfiltrated serosa type; 4) Gastric cancer COX-2 levels in diffuse growth pattern were significantly higher than those in mass or nest growth pattern; 5) LN COX-2 levels were significantly higher in positive LN metastasis than in negative LN metastasis. Conclusions: COX- 2 mRNA expressions in gastric cancer are significantly higher, and their expressions are related to biological behavior of gastric cancer; COX-2 mRNA expressions in LN may have some value in predicting LN metastasis of gastric cancer.
