摘要
目的针对发现的一种血管内皮生长因子变异基因(h′VEGF),利用计算机软件模拟预测其空间结构,并对其功能进行初步探讨。方法将pCD-hVEGF165及pCD-h′VEGF165重组真核表达载体导入CHO细胞,收集培养上清,进行ELISA分析、血管通透性实验(Miles实验)、内皮细胞增殖实验(MTT法)。结果h′VEGF165能使豚鼠的血管通透性增高,并促进ECV-304细胞增殖,但ELISA实验结果显示h′VEGF165与抗hVEGF165单抗未发生抗原抗体反应。结论h′VEGF165与hVEGF165在空间结构上可能存在差异,但现有的实验结果未见两者在功能上的明显差异。
Objective To study the relationship between the function and structure of h' VEGF165. Methods After structure prediction with computer, the eukaryotic expression vector pCD- h'VEGF 165 and pCD-hVEGF165 were transferred into CHO cells. The supernatant was collected and measured by ELISA assay. Afterward, vascular permeability enhancement assay, endothelial cell proliferation assay were applied to detect the biological activity of this new protein. Results The h'VEGF165 enhanced the vascular permeability of guinea pig, and stimulated the proliferation of ECV-304 in vitro, but it did not react with the coated mono-clone antibody-anti- hVEGF4-65. Conclusion The h'VEGF165 and the hVEGF165 have the same function despite some structural differences.
