HPLC测定荔枝不同器官中内源激素流程的优化

An optimized HPLC procedure for analyzing endogenous hormones in different organs of litchi

为了探讨内源激素的提取分离和分析测定的优化流程。试验在前人研究的基础上,以荔枝根、叶、果实为材料,针对荔枝样品富含多酚类和色素等杂质的特点,借鉴酶联免疫法(ELISA)中植物激素提取分离方法,对以往高效液相色谱(HPLC)分析荔枝内源激素的前处理方法进行了改进,配合选择适宜的HPLC色谱条件,摸索出荔枝不同器官中内源激素分析测定的优化流程。结果显示该流程可以联合测定IAA、GA3、ABA和ZT(回收率分别为78.52%、82.36%、79.83%和78.57%),且保留时间稳定,试验重现性好。整个提取分离、分析测定流程简便、快速、准确,适合荔枝材料内源激素的分析测定。

This paper aims to find out an optimal procedure for isolating and analyzing endogenous hormone. It was carried out on the basis of former reported methods, with roots, leaves and fruit as materials. To deal with the existence of large amount of polyphenols, pigments and other impurities, referred to the isolation procedure of Enzyme Linked Immunosorbent Assay （ELISA）, an improved High Performance Liquid Chromatography （HPLC） method suitable for analyzing endogenous hormone in different organs of litchi was established, which includes modified sample preparation procedures and chromatography profiles. Results showed that this procedure could analyze the IAA, GA3, ABA and ZT in all （recovery was 78.52%, 82,36%,79.83% and 78.57%, respectively）, with the sound retention time and repeat. It was proved that this procedure is simple, rapid and accurate for efficient determination of endogenous hormone in litchi samples.
