摘要 目的探讨三磷酸腺苷(ATP)对体外培养的小鼠肌源性干细胞(MDSC)增殖和细胞周期的影响。方法根据细胞贴壁能力的差异,采用差速贴壁法分离培养MDSC,应用MTT比色测定法和流式细胞仪(FCM)分析检测实验组和对照组细胞的增殖情况和细胞周期的变化。结果经ATP作用后,A490波长的光吸收值明显升高,处于S期的MDSC数量(S%)明显升高,G1%降低,增殖指数PrI值(S+G2M)%增高。结论ATP能促进小鼠MDSC增殖,这一作用可能是通过促进MDSC从G1期进入S期来实现的。 Obieetive To evaluate the effects of ATP on the proliferation and cell cycle of mouse muscle-derived stem cells (MDSC). Methods On the basis of the difference of cellular adherent ability, MDSC from primary cultures of mouse skeletal muscle were isolated and purified by the preplate technique. MTT eolorimetrie assay and flow eytometry (FCM) were used to detect the effects of eeU proliferation and the change of cell cycle. Results In experimental group, the photoabsortion values in A490 was increased significantly. FCM revealed that the number of MDSC in S phase ( S% ) was significantly increased, G1 % obviously decreased and the PrI (S + G2M) % enhanced when the cells were stimulated with ATP. Conclusion ATP enhances the proliferation of MDSC, which may be accomplished by accelerating the transition from G1 to S.
