肌肉移植后终板区神经源性聚集蛋白的分布变化及意义

摘要 目的明确大鼠股薄肌游离移植后终板区神经源性聚集蛋白(agrin)的分布变化规律。方法以免疫组织化学和Western blot的方法定性和定量的检测正常肌肉和移植术后大鼠股薄肌终板区神经源性agrin的分布变化。结果在正常神经支配肌肉的运动终板区表达低水平的神经源性agrin,肌肉移植后早期神经源性agrin明显减少,但并未完全消失,术后10周起逐渐恢复,到30周时仍低于对照组的水平(P<0.05)。结论神经源性agrin的分布变化可能是终板重建延迟以及重建达不到术前水平的重要影响因素,由此也间接影响到移植肌肉功能的恢复。 Objective To study the changes of distribution of neural agrin at the neuromuscular junction following musde free transfer, Methods The distribution of neural agrin was detected by immunohistology (EnVisiou method) and Western blot in the normal gracilis muscle and transfer muscle at different postoperative periods. Results There was a little anti-neural agrin staining on the endplate in normal gracilis muscle and then a greatly overall decrease during the early period of muscle transfer but not entirely absent. At 10th week, the expression of neural agrin recovered gradually, but failed to return to the presurgical state tested by Western blot method even at 30th week after muscle transfer. Conclusion The decline in AChR number, a simpler morphology and delay reconstruction of endplate and persistent weakness of muscle following free transfer may be attributed to these changes of distribution of neural agrin.
