摘要
目的:观察不同运动强度训练后,大鼠肾脏血管紧张素ⅡAT1受体表达的变化,为运动对肾脏内分泌功能影响的研究提供形态学依据。方法:健康雄性SD大鼠80只分为4组,实施不同强度运动训练后,采用免疫组织化学法和计算机图像分析技术,观察分析肾脏血管紧张素ⅡAT1受体表达的分布。结果:肾脏血管紧张素ⅡAT1受体主要分布在肾小球、近曲小管和远曲小管,以远曲小管表达最为丰富。小强度运动训练后,肾脏血管紧张素ⅡAT1受体免疫反应变化不明显;中等强度训练运动后,肾脏血管紧张素ⅡAT1受体免疫反应明显减弱;大强度运动训练后,肾脏血管紧张素ⅡAT1受体免疫反应明显增强。结论:小强度运动训练对肾脏血管紧张素ⅡAT1受体表达影响不明显;中等强度运动训练使肾脏血管紧张素ⅡAT1受体表达下调,以适应运动应激;大强度运动使肾脏血管紧张素ⅡAT1受体表达上调,提示大强度运动可能有导致肾脏损害的趋势。
Objective In order to provide a newer theoretical and morphologic basis for studying the effects of exercise on renal endocrine function, we investigated the changes in expression of renal angiotensin Ⅱ type 1 receptor in rats following exercise with different intensities. Methods Eighty healthy male SD rats were trained with different intensities and served as animal model for this study. The angiotensin Ⅱ type 1 receptor expression in kidney were observed using immunohistochemistry and computer image analysis technique. Results The renal angiotensin Ⅱ type 1 receptor mainly located in renal glomerulus, proximal convoluted tubule and distal convoluted tubule, especially in distal convoluted tubule. Immunoreaction of angiotensin Ⅱ type 1 receptor in kidney changed insignificantly after low intensity exercise, decreased remarkably after the moderate intensity exercise, and increased significantly after high intensity exercise. Conclusion Moderate intensity exercise down regulate the expression of the renal angiotensin Ⅱ type 1 receptor and thus promote the adaptability of kidney to exercise, whereas high intensity exercise may be potentially harmful to renal function.
