内皮缩血管肽1和促肾上腺皮质激素对体外培养的正常人黑素细胞增殖的影响 被引量:7

The effects of endothelin-1 and ACTH on the prolifertation of cultured normal human melanocytes

摘要 目的了解内皮缩血管肽1及促肾上腺皮质激素(ACTH)对体外培养的正常人黑素细胞增殖的影响。方法采用体外黑素细胞培养技术、四甲基偶氮唑蓝比色法(MTT)研究不同浓度内皮缩血管肽1、ACTH对黑素细胞增殖的影响。结果从0.1 ̄1000nmol/L浓度的内皮缩血管肽1均可不同程度地促进正常人黑素细胞的增殖,低浓度0.1nmol/L内皮缩血管肽1具有很好的促增殖作用。10-13 ̄10-9mol/L的ACTH也可不同程度地促进正常人黑素细胞的增殖,与对照组相比,P值均<0.05,其中又以高浓度10-9mol/LACTH促增殖能力最强,10-12mol/L、10-11mol/L、10-10mol/L3个浓度间差异无统计学意义。结论内皮缩血管肽1及ACTH具有促进体外培养的正常人黑素细胞增殖的作用,较低浓度内皮缩血管肽1即具有很好的促增殖作用,正常人生理水平(10-12 ̄10-11mol/L)及更高浓度ACTH均有较强的促增殖作用。 Objective To study the effects of endothelin-1 ( ET-1 ) and adrenocorticotropic hormone ( ACTH ) on the proliferation of cultured normal human melanocytes. Methods Normal human melanocytes were obtained from the foreskins of healthy men, and cultured in minimum essential medium ( MEM ) with 10% bovine serum. The cultured melanocytes of the 3rd passage were treated with various concentrations of ET-1 and ACTH. Cell proliferation was assessed by the MTT method. Results ET-1 (0.1 - 1 000 nmol/L ) promoted the proliferation of cultured human melanocytes. ACTH ( 10^-13 ~10^-9mol/L ) also promoted the proliferation of cultured human melanocytes ( P 〈 0.05 ), with 10^-9 mol/L being the most efficient concentration. There was no significant difference in the effect of ACTH at the concentrations of 10^-12 mol/L, 10^-11 mol/L and 10^-10mol/L. Conclusions ET-1 and ACTH could promote the proliferation of melanocytes, which occurs even at low concentrations of ET-1. ACTH, at physiological ( 10^-12 and 10^-11 mol/L ) or higher concentrations could also promote the proliferation of melanocytes. Therefore, these two factors might be utilized in non-carcinogen culture and transplantation of melanocytes as well as direct treatment of vitiligo in the future.
