摘要
血管生成素样蛋白(Angptl)3和Angptl4均是血管内皮生长因子家族成员。Angptl3在肝脏特异表达,而Angptl4主要在脂肪和肝脏表达,在心、肾等组织也有表达。Angptl3和Angptl4均可以通过抑制脂蛋白脂肪酶活性而调节脂质代谢,但在不同的营养状态下发挥作用。在胰岛素抵抗或胰岛素分泌缺陷时Angptl3的表达和血浆Angptl3水平明显升高,而血浆Angptl4水平在2型糖尿病患者是下降的。动物实验中给予外源性Angptl4可明显降低血糖浓度、改善机体葡萄糖耐量,提示它们与胰岛素抵抗有重要关系。
Angiopoietin-like protein 3(Angptl3) and Angiopoietin-like protein 4 (Angptl4) are two members of vascular endothelial growth factors family. Angptl3 is only expressed in liver, while Angptl4 is expressed predominantly in adipose tissue and liver, with detectable expression in several other tissues such as heart and kidney. Both Angptl3 and Angptl4 can regulate hpid metabolism by inhibiting the action of lipoprotein hpase during different nutritional states. Angptl3 is significantly increased in insulin-deficient and insulin-resistant states, but plasma Angptl4 is lower in patients with type 2 diabetes than those in healthy subjects. Angptl4 significantly decreases blood glucose and improves glucose tolerance in vivo. It suggests there is an important correlation between Angptl3, Angptl4 and insulin resistance.
