摘要
目的研究急性坏死性胰腺炎(ANP)肺组织中Toll样受体2、4(TLR2、4)mRNA的表达及其意义。方法采用逆行胰胆管注射牛磺胆酸钠(TAC)诱导ANP肺损伤动物模型,并分为假手术组、胰腺炎组和氯喹阻断组。计算肺组织学分值和肺损伤指数以评价肺损伤程度,荧光实时定量-PCR方法检测不同组不同时间点肺组织TLR2和TLR4mRNA表达变化。结果与假手术组比较,胰腺炎组大鼠3h时肺组织TLR2、4mRNA表达开始增高,12h时达到峰值(P<0.05),同时,肺损伤加重,肺组织TNF-α浓度升高,NO浓度逐渐降低;给予氯喹治疗后,TLR2、4mRNA表达降低,肺损伤程度减轻,肺组织TNF-α浓度降低,NO浓度显著升高。结论ANP时,肺组织内TLR2和TLR4的基因表达上调,其升高同肺组织损伤呈正相关,在ANP肺损伤的发生、发展中起一定作用。
Objective To investigate changes and potential roIes of lung expression of Toil-like receptor 2 and 4 gene in acute lung injury complicated by acute hemorrhage necrotizing pancreatitis (AHNP) in rats. Methods Fifty-six SD male rats were randomly divided into Sham operation group(n - 8), AHNP group(n 24) and Chloroquine inhibiteded group(n = 24). The lungs were dissected for lung histological scores, and bronchoalveolar lavages were harvested for lung injury index. TLR2/4 mRNA expression in lungs were measured by RT-PCR. Results Low level of TLR2/4 mRNA was detected in lungs of sham-operation group, and was found to be markedly increased at 3 h in AHNP group, peaking at 12 h. In AHNP group, lung injuries were aggravated, TNF-α level in lungs was increased and NO level was decreased compared with sham-operation group(P 〈 0.05). Treatment with CQ effectively inhibited TLR2/4mRNA expression at 3, 6 and 12 h (P 〈 0. 05) and relieved lung injuries, where TNF-α level in lungs decreased and NO level increased significantly (P 〈 0. 05). Conclusions These data suggest that expression of TLR2/4mRNA is increased in AHNP lungs. Upregulation of TLR2/4mRNA expression in lungs may be involved in the genesis and development of acute lung injury complicated by AHNP.
