贲门癌高发区贲门癌组织SLC5A8基因甲基化检测 被引量:4

摘要 目的:探讨SLC5A8基因甲基化在贲门癌变过程中的作用。方法:采用甲基化特异性PCR法检测42例贲门腺癌及15例癌旁组织SLC5A8基因的甲基化,采用RT-PCR方法检测42例贲门癌组织SLC5A8基因表达水平。结果:42例GCA组织中有18例(43%)SLC5A8基因甲基化阳性。15例癌旁贲门组织中有2例(13%)甲基化阳性。癌组织的甲基化阳性率(43%)显著高于癌旁正常组织(13%)(P<0.01)。贲门癌组织SLC5A8基因甲基化状况与肿瘤分化程度,肿瘤体积,淋巴结转移情况和远处转移情况等临床病理因素均无相关性。18例SLC5A8基因甲基化阳性标本中仅有3例(17%)检测到SLC5A8基因的表达;而24例SLC5A8基因甲基化阴性标本中有10例(42%)检测到SLC5A8基因的表达;SLC5A8基因甲基化阳性的贲门癌组织中SLC5A8基因表达率显著低于甲基化阴性的贲门癌组织的发生率(P<0.05)。结论:SLC5A8基因甲基化是GCA癌变过程中的重要分子事件,是SLC5A8基因失活的重要机制之一。 Aim: To investigate the methylation status of SLC5A8 gene in carcinogenesis of gastric cardia adenocarcinoma (GCA). Methods: Methylation-specific PCR (MSP) was used to analyze the status of SLC5A8 gene methylation in 42 cases of GCA and 15 cases of adjacent gastric cardiac tissue. SLC5A8 gene expression level in 42 cases of GCA was detected with RT-PCR method. Results: In 42 cases of GCA, the methylation rate of SLC5A8 gene in tumor tissue was 43% (18/42) , the methylation rate in adjacent cardiac tissue was 13% (2/15). The methylation rate in tumor tissue was remarkably higher than that in adjacent tissue (P 〈 0. 01 ). No significant association was found between the methylation status of SLC5A8 gene and tumor size, differentiation, nodal stage and metastasis (P 〉0.05 ). SLC5A8 gene expression was detected in 3 of 18 cases of ( 17% ) SLC5A8 methylation-positive GCA and 10 of 24 cases of (42%) SLC5A8 methylation- negative GCA. SLC5 A8 gene expression rate in SLC5 A8 methylation-positive GCA was lower thau that in SLC5 A8 methylation-negative GCA(P 〈0.05 ). Conclusion: The aberrant methylation of SLC5A8 gene is an important mechanism of SLC5A8 gene inactivation in GCA, and may play an important role in carcinogensis of GCA.
