摘要
目的探讨内皮细胞特异性分子-1(ESM-1)在转染MEK基因的人脐静脉内皮细胞中的表达,探讨ESM-1的表达是否受分裂原激活的蛋白激酶(MAPK)信号转导通路的调节。方法脂质体转染高活性的MEK基因入ECV304细胞,Westernblot检测ESM-1在ECV304细胞中的表达。结果成功获得稳定转染高活性MEK基因的细胞株,并检测到胞外信号调节的激酶(ERKs)的表达量在二株细胞中没有变化,而pERK-2和ESM-1在转染MEK基因的细胞中表达高于未转染的细胞。结论ESM-1可能处于ERK-2的下游,其表达可能受MAPK信号转导通路的调节。
Objective To transfect the recombinant MEK( mitogen-activated protein kinase/external-signal regulated kinase) gene of high activity into endothelial cells and examine the expression quantity of ESM-1 in transfected cells. And to study the relation between ESM-1 and MAPK (mitogen-activated protein kinase)signal transduction and tumor metastasis. Methods We transfected the recombinant MEK gene into human umbilic vein endothelial ceils ECV304, and western blot was used to examine the change of ESM-1 expression in cells transfected with recombinant MEK gene of different activities. Results After examining the ceils by western blot, we found that the cells transfected with the high activity type recombinant MEK gene expressed pERK-2 (external-signal regulated kinase-2) and ESM-1 more than ECV304 cells. Conclusion ESM-1 may be at the downstream of ERK-2 and be regulated by MAPK signal transduction pathway.
