摘要 目的探讨抑癌基因PTEN在子宫内膜癌中的表达及与子宫内膜癌临床、病理等因素的相关性。方法运用免疫组织化学(SP法)检测10例增生期子宫内膜、28例子宫内膜增殖症、8例不典型增生和54例子宫内膜癌组织中PTEN蛋白的表达情况,并比较PTEN蛋白表达与内膜癌临床、病理等因素之间的相关性。结果PTEN蛋白在增生期子宫内膜、子宫内膜增殖症、不典型增生和子宫内膜癌中阳性表达率呈下降趋势,分别为100%(10/10)、89·29%(25/28)、87·5%(7/8)和55·56%(30/54),在子宫内膜癌中的表达率显著低于增生子宫内膜(P=0·0087)和子宫内膜增殖症(P=0·0022),PTEN蛋白表达与内膜癌组织学类型、临床分期、组织分化有关。结论PTEN与子宫内膜癌发生、发展密切相关,与子宫内膜癌的组织学类型、临床分期、组织分化密切相关,PTEN有望成为内膜癌治疗的靶蛋白及判定预后的标记物。 Objective To elucidate the relationship between PTEN and the arising and development of endometrial cancer by studying the expression of them in the endometfial cancer clinical pathological factors. Methods We studied the expression of PTEN in the following samples including ten proliferative phase endometrium, twenty-eight endometrium hyperplasia, eight atypical hyperplasia, fifty-four endometrial cancer tissue by the immunohistochemicalanalysis( streptavidin-biotin peroxidase method) . We also compared the relationship between the variety of expression of PTEN and endometrial cancer clinical pathological factors. Results It was declining among proliferative phase endometrium, endometrium hyperplasia and endometrial cancer tissue, and the positive rate was 100% (10/ 10), 89. 29% (25/28), 87.5% (7/8), 55.56% (30/54) respectively. The positive rate of PTEN in endometrial cancer tissue was significantly lower than those in proliferative phase endometrium ( P 〈 0. 05 ) and endometrium hyperplasia ( P 〈 0. 05 ). It had relationship between expression of PTEN and endometrial cancer histological types, clinical stages and histological differentiation. Conclusion PTEN has higher relationship with the arising and development of endometrial cancer, also with its histological types, surgerical-clinical stages, pathological Classifications, while they may be target proteins in the treatment of endometrial.
