外源性雌激素对去卵巢大鼠抗氧化能力及海马神经元线粒体的影响 被引量:8

摘要 目的用去卵巢大鼠作为雌性更年期的动物模型,探讨补充外源性雌激素对血清抗氧化能力、海马神经元线粒体形态结构的影响及与凋亡的关系。方法用HE染色和电子显微镜观察海马神经元细胞形态和线粒体超微结构的变化,用荧光显微镜检测海马神经元的凋亡,用生化法和RIA测定血清中MDA、SOD和E2等指标。结果未补E2组海马神经元线粒体出现损伤,补充大剂量E2组,其形态基本正常;血清中MDA、SOD与E2浓度呈负相关;未补充E2组海马神经元凋亡明显,随着补充剂量增加,凋亡逐渐减少。结论外源性E2对海马区神经元的保护作用,可能与血清抗氧化能力提高,减少线粒体损伤和海马神经元凋亡有关。 Objective This experiment used ovariectomized rats as models of female climacteric state, for studying the exogenous estrogen affected on antioxidation capacity in sermn, structure of neuron mitochondria and apoptosis of hippocampus neuron in ovariectomized rats. Methods Observing the cell structure changes by hematoxylin-eosin stain(HE stain), the cell ultra microstructure changes by electron microscope(EM), neuronal apoptosis by fluorescence microscope; determining the concentration of malondialdehyde(MDA), estrogenic hormone(E2 ) and superoxide dismutase(SOD) with biochemistry method or radioimmunoassay(RIA). Results Mitochondrial damages were found in hippocampal neurons of ovariectomized rats, but not be found in control group;the concentration of MDA and E2 in blood sermn was inverse correlation, SOD and E2 in blood sermn was also negative correlation; the numbers of hippocampus neuron apoptosis reduced consistented with the increase of E2. Conclusion The supplement of exogenous estrogen maybe reduce body peroxidation injury to protect hippocampal neurons.
