摘要
目的探讨中老年人群肠脂肪酸结合蛋白(IFABP)基因多态性频率分布与血清血脂水平的关系。方法中老年男性217例,年龄56.2±10.1岁;女性252例,年龄55.4±12.6岁。利用聚合酶链反应(PCR)、HhaI内切酶酶切、基因测序等技术检测各组IFABP基因型,同时检测血清总胆固醇(TC)、三酯酰甘油(TG)、高密度脂蛋白胆固醇(HDLC)、低密度脂蛋白胆固醇(LDLC)、载脂蛋白AⅠ(apoAⅠ)、载脂蛋白B(apoB)等指标。结果中老年人群54A/TIFABP基因型频率分布,男性组:A/A0.48,A/T0.42,T/T0.10;女性组:A/A0.44,A/T0.46,T/T0.10。突变型54T等位基因频率分别为男性组0.31,女性组0.33(P>0.05)。男性组54T等位基携带者血清LDLC、apoB高于54A等位基因携带者(分别为2.38±0.63vs2.21±0.57mmol/L,P<0.05;0.67±0.21vs0.61±0.18g/L,P<0.05);女性组54T等位基携带者血清TC、LDLC高于54A等位基因携带者(分别为4.46±0.89vs4.21±1.00mmol/L,P<0.05;2.39±0.47vs2.22±0.75mmol/L,P<0.05)。结论在中老年人群中,IFABP基因多态性频率分布与性别无关;54TIFABP基因携带者有高血脂特征。
Objective To investigate the association of frequency of encoding 54Ala/Thr (A/T) single nucleotide polymorphism (SNP) in the intestinal fatty acid binding protein (IFABP) with serum lipids in middle aged and senile population. Methods The subjects under investigation included 217 middle aged and senile men with mean age of 56. 2±10. 1 years, and 252 middle aged and senile women with mean age of 55.4±12. 6 years. The Ala54 Thr allelic variant was detected by polymerase chain reaction, Hha I digestion, and sequen cing. TC, TG, HDL-C, LDL-C, apoAI and apoB in fasting serum were determined with biochemical technique. Results The genotype frequencies of A/A, A/T, T/T were 0. 48, 0. 42 0. 10 in men, 0. 44, 0. 46, 0. 10 in women, respectively. The allelic frequency of point mutant 54Thr in IFABP gene was 0. 31 in men, 0. 33 in women, respectively. There was no difference of the allelic frequency of 54T between men and women(X^2=0. 47, P〉0. 05). The LDL-C and apoB concentrations in fasting serum in men with 54T allele were signifi- cantly higher than those with 54A allele (2. 38±0. 63 vs 2. 21±0. 57mmol/L, P〈0. 05; 0. 67±0. 21 vs O. 61±0. 18g/L, P〈0. 05). The TC and LDL-C concentrations in fasting serum in women with 54T allele were significantly higher than those with 54A allele(4. 46±0. 89 vs 4.21 ±1. 00mmol/l., P〈0.05; 2.39±0.47 vs 2.22±0. 75mmol/l., P〈0.05). Conclusions In middle aged and senile population, the frequency of encoding 54Ala/Thr polymorphism in IFABP gene is not related with gender, but related with the high lipoprotein profile.
