摘要 目的建立人食管癌细胞系为潮汕沿海地区食管癌研究提供实验模型。方法采用胶原酶消化法建立人食管癌细胞系CSEC。研究培养细胞的形态特点、生长特征、对Sc id小鼠的致瘤性、细胞遗传学特点及肿瘤相关标志物(AE 3、p53)的表达。结果细胞系CSEC在体外持续稳定生长超过13个月,已传至80代以上,群体倍增时间为39.6小时。CSEC细胞具有鳞状细胞的形态和结构特点,胞浆富含张力原纤维束,细胞间可见桥粒。Sc id小鼠移植瘤的组织学形态与患者的原发肿瘤一致。细胞系CSEC呈近四倍体核型,染色体结构改变常见而且复杂。肿瘤相关标志物(AE 3、p53)呈强阳性表达。结论人食管癌细胞系CSEC是一分化较高且生物学特性稳定的鳞状细胞癌细胞系,可为食管癌的发病机制和治疗研究提供有价值的实验模型。 Objective To establish a new human esophageal carcinoma cell line and to provide a experiment model for research on esophageal carcinoma in Chaoshan littoral a high-incidence area of esophageal carcinoma. Methods The esophageal carcinoma cell line CSEC was established by collagenase digestion methods. The biological characters of the cultured cells were investigated, including morphology, ultrastructure, growth kinetic features, tumorigenicity to Scid mice, cytogenetic features and expression of tumor associated markers (AE3,p53). Results The cell line CSEC grew continuously in vitro with a doubling time of 39.6 hours and was passaged over 80 times over 13 months. The CSEC cells possessed the morphology and structure features of squamous cell with tonofilaments and desmosomes revealed by electron microscopy. The CSEC cell line showed tumorigenicity to Scid mice and the histologic features of the tumors developed were similar to the origin tumor from which the cell line derived. The cytogenetic analysis demonstrated hypotetraploid karyotypes. Chromosome structure aberrations were common and complex. Immunohistochemical staining showed that CSEC cells expressed AE3 and p53. Conclusions The CSEC cell line is a well-differentiated esophageal squamous cell carcinoma cell line with stable biological characters. It could provide a useful model for the pathogenesis and therapeutic study for esophageal squamous cell carcinoma.
