大鼠坐骨神经横断后脊髓、背根神经节及坐骨神经 Slit 2表达的变化 被引量:5

Changes of Slit 2 expression in spinal cord, dorsal root ganglion and sciatic nerve after sciatic nerve transection in rats

摘要 目的:观察 Slit 2在大鼠坐骨神经横断模型中的表达变化,为进一步研究 Slit/Robo 在周围神经再生中的作用提供实验依据。方法:SD 大鼠坐骨神经横断后用原位杂交及免疫组织化学方法检测 Slit 2在脊髓、背根神经节(DRG)和横断神经近、远端内的表达变化,图像分析方法测定阳性细胞数及平均积分光密度值。结果:正常脊髓前角运动神经元、DRG 和神经干内 Slit 2有一定的基础表达。损伤后近端神经胶质瘤、神经远端 Slit 2表达增高;DRG 内的 Slit 2表达呈现一定的时间变化,7 d 为表达高峰,14 d 下降。结论:Slit 2存在于正常成年大鼠周围神经系统,损伤可致其表达改变,Slit 2可能在周围神经再生中发挥重要作用。 Objective: To investigate the changes of Slit 2 expression in rats with sciatic nerve transection, so as to provide evidence for the role of Slit/Robo in regeneration of peripheral nerve. Methods: The sciatic nerve was transected in Sprague-Dawley rats, and the changes of Slit 2 expression were detected by in situ hybridization and immunohistochemical methods in related spinal cord, dorsal root ganglion (DRG), distal and proximal stump of transected sciatic nerve at 1 d, 3 d, 7 d, 14 d and 21 d, respectively. Then the number of positive cells and average integral optical density value (AIODV) were determined with image analysis. Results: Slit 2 was expressed in normal anterior horn of spinal cord, DRG and nerve trunk; the expression was up-regulated in the distal stumps and glioma after sciatic nerve transection. However, changes of Slit 2 expression in DRG peaked on d 7 then decreased on d 14. Conclusion.. Slit 2 is expressed in the peripheral nerve system of adult rat and the expression can be influenced by injury, suggesting that Slit 2 may play an important role in the regeneration of peripheral nerve after injury.
