几种海水经济贝类DNA提取方法探讨

Methods for DNA Extraction from Shellfishes

以缢蛏、美人蛏、文蛤、扇贝和鲍鱼等几种经济贝类为研究对象,分别用苯酚／氯仿／异戊醇法,CTAB法,SDS法等不同方法提取其基因组DNA,寻找适宜于研究对象基因组DNA提取的方法,结果表明,3种方法均可以从目标材料中提取到DNA,但苯酚／氯仿／异戊醇法无论从数量和质量上都不理想,而SDS和CTAB可根据具体情况选择使用。即对纯度要求不高时均可选用SDS 法,但为了分子生物学研究的应用,需要较高纯度,则缢蛏、美人蛏、文蛤和扇贝等瓣腮纲贝类推荐用SDS法,而鲍鱼等腹足纲贝类推荐用CTAB法。

We adopted the phenol-chloroform-isopentanol, CTAB and SDS methods to extract DNA from Sinononacula constricta, Meretix linnaeus, Chlamys nobilis, Haliotis diversicotor and other shellfishes. Results showed that the above methods can help us to extract DNA, but SDS is the best one for all objected shellfishes when we want to have high yield. However, when we want to have DNA of high purity, SDS is suitable only for Sinononacula constricta, Meretix linnaeus and Chlamys nobilis, and CTAB is the best one for Haliotis diversicolor.
