顺铂和电子线照射对人肺癌细胞凋亡的影响

摘要 探讨单纯照射和单纯顺铂以及照射联合顺铂对肺癌细胞凋亡的影响。应用细胞培养技术,分别采用单纯照射,单纯顺铂,照射联合顺铂用于人肺癌细胞SPC -A4细胞,通过荧光染色法、DNA凝胶电泳分析等检测方法观察其诱导凋亡的差异。并进行量化以比较3种方法诱导细胞凋亡效果差异的程度。结果表明,顺铂组、照射组、顺铂联合照射组,DNA琼脂糖凝胶电泳及裂解显示凋亡发生。荧光染色法细胞凋亡的定性观察、定量分析表明,顺铂诱导的细胞凋亡呈时间依赖性,照射组中以6Gy和8Gy照射12h的凋亡率最高,在2 4h时凋亡率反而下降,顺铂联合照射与单纯照射及单纯顺铂相比,在对应时间段相比凋亡率前者明显高于后两者(P <0 .0 0 1)。结果表明,单纯顺铂和单纯照射及顺铂联合照射均能诱导肺癌细胞凋亡,顺铂联合照射诱导肺癌细胞的凋亡率明显高于单纯顺铂及单纯照射。 To study how to enhance the apoptosis rote of lung cancer cell, to find more effective treatment for lung cancer, the effect of lung cancer cell apoptosis induced by electron rays or cisplatin or combination of cisplatin with electron rays was studied, Fluorescent stain and DNA agarose gel electrophoresis technique were used and the formation of cellular apoptosis was observed. The effects of three methods on cellular apoptosis were compared. Result showed that the cisplatin inducing apoptosis, apoptotic peak reached after radiation 12 hours at the dose of 6Gy and 8Gy, apoptotic rate decreased after radiation at the dose of 10Gy, the combined way achieved better effect of apoptosis than cisplatin only or radiation in the same time phase with a significance between them( P 〈 0.001). Apoptosis of lung cancer cell can be induced by combination of cisplatin and electron rays or only cisplatin or electron rays. The combined way achieved better effect of apoptosis than cisplatin only or electron rays. Cisplatin enhancs sensibility of electron rays.
