摘要
目的:探讨鼠巨细胞病毒(MCMV)感染的不同时期MCMV M83基因mRNA在小鼠睾丸中的表达及与睾丸病变程度的相关性。方法:采用地高辛标记的MCMV M83mRNA寡核苷酸探针对接种MCMV后不同时期的小鼠睾丸组织进行原位杂交(ISH)检测。将ISH阳性杂交信号的平均光吸度(A)值表示睾丸的MCMV负荷量,分析MCMV M83mRNA在感染睾丸中的表达及其负荷量与睾丸病变程度的关系。结果:接种病毒后各时期的睾丸组织中均有MCMV M83mRNA表达,ISH阳性杂交信号主要定位在间质细胞胞浆内。感染d9的平均A值最高(实验组组间比较,P<0.001)。感染不同时期的生精上皮及间质细胞呈现不同程度的病理性炎性改变,d6-9最为严重。结论:睾丸MCMV感染时,MCMV M83基因的mRNA水平与睾丸感染病变程度相关;提示:对MCMV M83mRNA动态检测可估计感染组织的预后。
Objective: To investigate the expression of murine cytomegalovirus (MCMV) M83 mRNA in mouse testes at the different periods of MCMV infection and its relation with the severity of testicular lesions. Methods: By using digoxin-labeled MCMV mRNA oligonucleotide probe, the mouse testicular tissues at the different periods following MCMV inoculation were detected by in situ hybridization, The average absorbency (A) of ISH positive signals represented the resticular MCMV load. The expression of MCMV M83 mRNA in the infected testes and its relation with the severity of testicular lesions was analyzed. Results: At different periods after MCMV inoculation, the expression of MCMV M83 mRNA was detectable in the testicular tissues. The ISH positive signals were mainly located in cytoplasm of the testicular interstitial cells. The average A values at d 9 after MCMV infection were the highest (comparision between the experimental groups, P〈0.001), There were pathological inflammatory changes to varying degrees in seminiferous epithelia and interstitial cells at the different periods of infection, especially during d 6 to d 9. Conclusion: In testicular MCMV infection, the levels of MCMV M83 mRNA was related with the severity of testicular lesions, suggesting the dynamic detection of MCMV M83 mRNA could evaluate the prognosis of MCMV infection.
