摘要
目的:探讨不同机械切口辅助孵化方式对小鼠胚胎体外发育的影响。方法:将678枚2-4-细胞期昆明白种小鼠胚胎随机分为A(183枚)、B(177枚)、C(168枚)组和对照组(150枚)。A、B、C组采用机械法在透明带上分别开“─”、“┼”、“┤”形状切口,对照组不作切口,比较各组的囊胚获得率和孵出率。结果:A、B、C组d5囊胚获得率分别为92.35%、92.09%和91.07%,与对照组的93.33%比较,无统计学差异(P>0.05);d6囊胚完全孵出率依次为75.00%、91.52%和90.57%,显著高于对照组的48.25%(P<0.001)。B、C组d6囊胚完全孵出率无统计学差异(P>0.05),但均显著高于A组(P≤0.001)。A组的辅助孵化时间(13.88±1.72s)显著小于B、C组(44.46±2.61s和53.73±3.29s)(P<0.05);且B、C组间也有显著差异,P<0.05。结论:不同机械切口的辅助孵化方式均能有效提高囊胚孵出率;“┼”和“┤”形状切口的辅助孵化效果显著优于单一切口者;“┤”较“┼”形状切口的辅助孵化方式更省时、安全,较“─”方式更有效,值得推广。
Ohjective: To evaluate the safety and efficiency of assisted hatching(AH) by different mechanic technique on mouse embryos in vitro development. Methods: A total of 678 KM BAI mouse embryos in 2-4- cell stage were randomly divided into group A, B, C and control group. ZP was made a "─ "', "┼" and "┤ "shaped slits respectively by different mechanic AH modes in group A, B, C in order to compare the rate ofblastocyst and hatching in four groups. Results: The rate ofblastocyst was not significantly different in three experimental groups and control group on d 5 (P〉0.05).Compared with the control group, the rate of completed hatching significantly increased in experimental groups on d 6 (P〈0.001).The rate of completed hatching in group B and C was not significantly different (P〉0.05), while significantly higher than that in group A respectively (P ≤ 0.001). The duration of AH in group C was significantly shorter than that in group B, but longer than that in group A (P〈0.05). Conclusion: Different mechanic AH modes were safety and efficiency for embryos in vitro development. The mode of "┤ " shaped slits was more safety and simple than those of" ┼" shaped slits, and efficiency than that of" ─" shaped slit. It may be one of the best AH methods for animal or human assisted reproduction technology.
