摘要 研究了在无溶剂体系中Α-糜蛋白酶催化N-BOC-L-苯丙氨酸乙酯与D-葡萄糖的酯交换反应,考察了水含量、反应时间、搅拌以及底物摩尔比等因素对酯交换反应的影响。结果表明是在6位伯羟基(6-OH)进行定位酰化。生成的氨基酸葡萄糖酯可通过HPLC-UV定量检测,最适条件下的反应转化率达到90%以上。LC-MS分离并初步定性分析,产物相对分子质量为445,与理论目标产物的一致。 The transesterifiction of N-BOC-L-phenylalanine ethyl ester with D-glucose catalyzect by α-cnymotryps,n in solvent-free system was studied. The effects of water activity or water content, reaction time, stirring on the transesterification were investigated. Results shown that D-glucose was selectively acylated at one primary hydroxyl (6-OH). The product could be tested qualitatively and quantitatively by HPLC-UV. The conversion was beyond 90% under the best conditions. The molecular weight of product is 445, which is corresponding to the theoretical product.
