酶法水解丝素蛋白最佳条件的研究 被引量:6

摘要 通过6种酶对家蚕丝素蛋白水解的试验表明,Alcalase 2.4L水解效果最佳,Flavorzyroe次之,并经正交试验确定Alealase对丝素水解的最佳条件为:浓度8%的丝素溶液中加入1%的酶,在pH值8、55℃条件下反应6h,水解度可达39.34%,回收率为76%。SDS-PAGE电泳实验证明,水解产物的分子量大致为6.2-7.8kDa。在此基础上,添加1.5%的Flavorzyme继续水解,在pH值7.50℃条件下反应3h,水解度为44.13%,回收率为59%。 In the paper six kinds of enzymes were used to deal with respectively silk fibroin protein of Bombyx mori. Alcalase 2.4 L was the best and Flavarzyme was the second .The results of orthogonal design showed that the optimum condition for basicity protease. Alcalase acting on silk fibrion was 8% substrate concentration adding 1% Alcalase, pH 8.55℃ and 6 hours, In such condition, the hydrolysis degree can reach 39.34 % and its rate of recovery reached 76%, The results of measuring the molecular weight of silk fibroin after being hydrolyzed were about 6.2 - 7.8 kDa by SDS-PAGE. On the based of that,we added 1.5% Flavorzyme going on hydrolyzing, in the condition of pH 7.50℃ ,reacting 3 hours, the hydrolysis degree was 44.13% and its rate of recovery reached 59%.
