Vectors Used in Genetic Engineering in Silkworm

摘要 基因工程分为将外源DNA整合到宿主染色体的种系转移和外源DNA不整合到宿主染色体的瞬时表达,用于家蚕基因工程的载体主要有两类,一类是基于转座子的载体:Minos转座子载体、Mosl元件载体和PiggyBac转座子载体。另一类是基于病毒的载体:杆状病毒载体S、indbis病毒载体和假型反转录病毒载体。家蚕种系转移主要采用piggyBac转座子载体,而外源基因的表达则主要采用杆状病毒载体和家蚕培养细胞组成的NPV-Bm表达系统,Sindbis病毒载体在家蚕RNA干涉的研究中显示出了诱人的前景。 Genetic engineering includes the gene transformation technique that exogenous DNA is integrated into the host come and the transient expression technique that exogenous DNA is not integrated into the host chromosome. Vectors used in genetic engineering in the silkworm are divided into two types. The first one is transposon-based vectors, such as Minos, Mosl element and PiggyBac vectors. The second is viral vectors, such as baculovirus, sindbis virus, Pseudotyped retroviral vectors. PiggyBac vectors are commonly used in gene trandomation technique. NPV-Bm express system is usually adopted in transient expression technique of exogenous gene. Sindbis virus vectors can become an important tool for RNAi study.
