摘要
目的建立肾石通颗粒中槲皮素的含量测定方法。方法采用高效液相色谱法Kromasil C18色谱柱(250mm×4,6mm),以甲醇-0.4%磷酸溶液(50:50)为流动相,检测波长:370nm,流速:1,0ml·min^-1,柱温:40℃。结果HPLC法可测定肾石通颗粒中槲皮素的含量,结果验证槲皮素在5.20-20.80mg·L^-1的浓度范围内峰面积与量呈良好的线性关系,回归方程为:A=3,858×10^5×C+8.094×10^3(r=0.99998,n=7);平均加样回收率可达98.9%(RSD=0.40%,n=9)。结论该法具有专属、可靠、灵敏度高、干扰小等特点,可作为肾石通颗粒中槲皮素的含量测定方法。
Aim To estalish a method for determining the conent of quercetin in shenshitong grainule. Methods HPLC method was used . The samples were separated by symmetry shield TM RP C18 column. The mobile phase is methanol-0.4% phosphoric acid (50: 50), The detection wavelength was 370 nm. Flow rate was 1.0 ml · min^-1 The column temperature was at 40℃. Results The linear ranges of quercetin is between 5.20 mg·L^-1 and 20.80 mg · L^-1. The average recovery is 98.9% ( RSD = 0.4%, n = 9). Conclusions The method is sensitive,rapid and accurate. It is suitable for determining shenshitong grainual.
