高效液相色谱法测定肾石通颗粒中槲皮素的含量

Determination of quercetin in shenshitong grainule by HPLC

目的建立肾石通颗粒中槲皮素的含量测定方法。方法采用高效液相色谱法Kromasil C18色谱柱（250mm×4，6mm），以甲醇-0．4％磷酸溶液（50：50）为流动相，检测波长：370nm，流速：1，0ml·min^-1，柱温：40℃。结果HPLC法可测定肾石通颗粒中槲皮素的含量，结果验证槲皮素在5．20-20．80mg·L^-1的浓度范围内峰面积与量呈良好的线性关系，回归方程为：A=3，858×10^5×C＋8．094×10^3（r=0．99998，n=7）；平均加样回收率可达98．9％（RSD=0．40％，n=9）。结论该法具有专属、可靠、灵敏度高、干扰小等特点，可作为肾石通颗粒中槲皮素的含量测定方法。

Aim To estalish a method for determining the conent of quercetin in shenshitong grainule. Methods HPLC method was used . The samples were separated by symmetry shield TM RP C18 column. The mobile phase is methanol-0.4% phosphoric acid （50： 50）, The detection wavelength was 370 nm. Flow rate was 1.0 ml · min^-1 The column temperature was at 40℃. Results The linear ranges of quercetin is between 5.20 mg·L^-1 and 20.80 mg · L^-1. The average recovery is 98.9% （ RSD = 0.4%, n = 9）. Conclusions The method is sensitive,rapid and accurate. It is suitable for determining shenshitong grainual.
