摘要
目的:采用高效液相色谱法测定中药材穿山龙中薯蓣皂苷元的含量。方法:高效液相色谱法,十八烷基硅烷键合硅胶为填充剂,Alltima C_(18)(150mm×4.6mm,5μm),甲醇-水(84:16)为流动相,流速:1mL·min^(-1),检测波长为203nm,柱温:25℃。结果:薯蓣皂苷元1.28~13.10μg范围内色谱响应线性关系良好,r=0.9995,回归方程为 Y=1.550×10~4X+396.7。仪器精密度的 RSD 为1.6%,方法重复性的 RSD 为0.94%。平均回收率为97.7%,RSD=0.60%(n=6)。结论:本法快速简便,重复性好,测定准确灵敏。
Objective: To develop an RP - HPLC method for the determination of diosgenin in Dioscorea nipponica Makino. Method:An Alltima C18 column( 150 mm ×4. 6 mm ,5 μm) was used with the mobile phase of methanol - water (84: 16) at 25 ℃, flow rate being 1.0 mL ·min 1 ,detecting wavelength being 203 nm. Results:The calibration curve was linear in the range of 1.28 - 13.10 μg of diosgenin with correlation coefficient of 0. 9995 and the regression equation as Y = 1. 550 × 10^4 X +396. 7. Diosgenin was steady in methanol within 5 days and the instrument precision RSD was 1.6% while the method precision RSD was 0. 94%. The average recovery was 97.7%, RSD = 0. 60% ( n = 6). Conclusion: The method is quick, accurate, sensitive, and simple with good repetition.
