毛细管电泳法测定虎杖药材中白藜芦醇苷、大黄素和大黄素甲醚的含量 被引量:7

摘要 目的:建立毛细管电泳法测定虎杖药材中活性成分白藜芦醇苷、大黄素和大黄素甲醚的含量。方法:采用 Agilent^(3D)CE高效毛细管电泳仪,未涂渍熔融石英毛细管(75μm×48.5cm,有效长度40.0cm),压力进样5.25Pa×3s,运行缓冲液为10mmol·L^(-1)硼酸钠溶液-甲醇(1:1),检测波长为220nm,运行电压30kV,温度25℃,内标为厚朴酚。结果:白藜芦醇苷、大黄素和大黄素甲醚浓度分别在20.5~410.8μg·mL^(-1)(r=0.9990),9.78~156.4μg·mL^(-1)(r=0.9998),12.13~121.3μg·mL^(-1)(r=0.9992)的范围内呈良好线性相关;平均回收率分别为98.9%,99.0%,96.9%,RSD 分别小于1.8%,2.1%,1.6%。结论:该方法简便快速、准确、重复性好,可用于虎杖中自藜芦醇苷、大黄素和大黄素甲醚的含量测定。 Objective: To determine the content of resveratrol - 3 - O - β - D - glucoside, emodin and rheochrysi- din in Rhizoma Polygoni Cuspidati by capillary zone electrophoresis. Method: Untreated fused silica capillary (48.5 cm ×75 μm,effective length is40 cm) was used. Pressure injection orS. 25 Pa ×3 s was employed,and the temperature was 25 ℃. The detection wavelength was set at 220 nm. The samples were analyzed with 10 mmol · L^-1 sodium borate - methanol ( 1 : 1 ) running buffer at 30 kV voltage. The internal standard substance was magnolol. Results: The calibration curve of resveratrol - 3 - O - β - D - glucoside, emodin and rheochrysidin showed good linearity over the range of 20. 5 -410. 8 μg · mL^-1 (r =0. 9990), 9.78 - 156. 4 μg · mL^-1 (r =0. 9998), 12. 13 - 121.3 μg· mL^-1z ( r = 0. 9992 ), respectively. The average recoveries of the method were 98.9% ,99.0% and 96. 9% with RSD less than 1.8% ,2. 1% and 1.6% ,respectively. Conclusion:The method is simple,rapid,accurate, reproducible and can be applied in the determination of resveratrol - 3 - O - β - D - glucoside, emodin and rheochrysidin in Rhizoma Polygoni Cuspidati
