摘要
目的:以环糊精及其衍生物为手性选择剂,通过优化缓冲液的浓度、酸度以及采用环糊精的种类和浓度以达到佐米曲坦的基线分离,并将建立方法应用于实际样品的质量控制中。方法:用20mmol·L^(-1)的磷酸二氢钾-磷酸缓冲液,pH=2.0,含有10mmol·L^(-1)羟丙基-β-环糊精(HP-β-CD)的运行液,分离电压为25kV,检测波长226nm。结果:佐米曲坦对映体的分离度达到3.1,本方法可以灵敏检出限量为0.5%的 R 构型光学异构体。结论:方法简便,快速,准确,可有效控制本品中R对映体杂质含量。
Objective:To establish HPCE methods for separating and determining the zolmitriptan enantiomers and to investigate the influence of relative factors, such as buffer concentration, pH, CDs types and their concentration, on the chiral separation of zolmitriptan. Methods:The buffer solution adopted was 20 mmo· L^-1 phosphate solution, pH =2. 0, with 10 mmol· L^-1HP - β - CD as chiral selector. Applied voltage was 25 kV, and the detection wavelength was 226 nm. Results: Under the optimum experiment conditions, the baseline separation of zolmitriptan enantiomers was obtained,with the resolution better than 3.1. Conclusion:The proposed method is simple, rapid and accurate and can be used for the quality control of the chiral purity.
