RP-HPLC法测定浙贝炮制品中主要生物碱的含量

RP-HPLC determination of peimine and peiminine in different processed products of Fritillaria thunbergii Miq.

目的:利用梯度洗脱、RP-HPLC 法同时测定浙贝母炮制品中的贝母素甲、贝母素乙。方法:采用 Xterra RP18色谱柱(3.9mm×150mm,5μm),流动相:乙腈-10mmol·L^(-1)碳酸氢铵(浓氨水调 pH10.10)系统,梯度洗脱,流速0.8mL·min^(-1),柱温:40℃,检测波长210nm。结果:在此条件下,贝母素甲、贝母素乙的进样量分别在1.09～4.36μg和1.04～4.16μg的范围内与峰面积呈线性关系;回收率分别为92.30%及94.50%,RSD 为1.1%及1.2%。结论:本法用于测定浙贝母炮制品中的贝母素甲、贝母素乙含量,简便、准确。

Objective：To develop a method for determination of peimine and peiminine in different processed products of Fritillaria thunbergii Miq. by RP - HPLC. Method： The HPLC system consisted of Xterra RP18 column （5 μm,3.9 mm × 150 mm）, acetonitrile- 10 mmol · L^-1NH4 HCO3 （adjusted to pH 10. 10 by ammonia solution） as mobile phase ,eluted in gradient mode ,flow rate ：0. 8 mL · min^-1 ,detected by UV detector. Result：The recoveries of peimine and peiminine were 92.30%, with RSD 1.1% and 94, 50%, with RSD 1.2% respectively. Conclusion：The method is appropriate for the determination of peimine and peiminine
