摘要 目的:建立检测聚乙二醇化促红细胞生成素(PEG-EPO)产品中残余聚乙二醇(mPEG_2-COOH)含量的方法。方法:样品经十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE)后,采用碘染色法进行染色,根据染色带深浅分析样品中 mPEG_2-COOH 的含量。结果:确定了测量 PEG-EPO 中检测 mPEG_2-COOH 含量的实验条件,在本实验条件下 mPEG_2-COOH 的检测下限约为5μg·mL^(-1)。结论:本法便捷、重复性好,适合该 mPEG_2-COOH 含量的检测要求。同时可运用于相关的聚乙二醇修饰蛋白质。 Objective :To establish a method to detect and determine the content of mPEG2 -COOH in the semimanufactured PEG - EPO, Methods: Electrophoresis the sample in SDS - PAGE, then treat the gels with iodine reagents, determine mPEG2 -COOH according to the dot degree. Results :The method was established, and the limit of detection of this method for mPEG2 - COOH is 5 μg · mL^- 1. Conclusion: The method is convenient, reproducible for detecting and determining the mPEG2 -COOH, and can be used in correlational PEG conjugated protein.
