摘要
目的:建立 HPLC 法测定毛叶钝果寄生中3种主要黄酮成分的含量。方法:采用 HPLC 法,色谱柱为 Zorbax SB-C_(18)柱(4.6mm×250mm,5μm),以乙腈-甲醇-1%乙酸溶液(18:13:69)为流动相,流速为0.8mL·min^(-1),检测波长254nm,测定毛叶钝果寄生中金丝桃苷、芦丁、广寄生苷的含量。结果:金丝桃苷、芦丁及广寄生苷的线性范围分别为16.72~836,19.68~984,19.84~992μg·mL^(-1),相关系数均为0.9998。加样回收率(n=5)分别为100.1%,100.2%,101.8%;RSD 分别为3.4%,3.5%,1.7%。毛叶钝果寄生样品中3种成分的平均含量(n=5)分别为0.206%,0.069%,0.051%;RSD 分别为3.1%,3.2%,2.7%。结论:该方法准确、重现性好,可作为毛叶钝果寄生药材的质量控制标准。
Objective :To establish an HPLC method for the determination of the contents of three main flavones in Taxillus nigrans (Hance)Danser. Methods:Contents of hyperin ,rutin ,guajeverin in Taxillus nigrans was measured by HPLC [ a column of Zorbax SB -C18 (4. 6 mm × 250 mm ,5μm) ,with mobile phase of acetonitrile -methanol - 1% acetic acid( 18: 13: 69) ,flow rate 0. 8 mL · min^-1 ,detection wavelength of 254 nm]. Results:The calibration curves showed good linearity in the range of 16. 72 -836 μg · mL^-1 (hyperin) ,19. 68 -984 μg ·mL^-1 (rutin) and 19. 84-992 μg · mL^-1(guajeverin) ;correlation coefficients were all 0. 9998. The average recoveries( n = 5 )were 100. 1% ,100. 2% and 101.8% ;RSD were 3.4% ,3.5% and 1.7% ,respectively. The average contents(n =5)of the three components in Taxillus nigrans were 0. 206% ,0. 069% and 0. 051% ,and RSD were 3. 1% ,3.2% and 2. 7%. Conclusion:An accurate and reproducible method can be used for the quality control of Chinese medicine Taxillus nigrans (Hance) Danser.
