摘要 目的:建立迪可颗粒中粉防己碱和防己诺林碱含量的测定方法。方法:采用高效液相色谱法,色谱柱为 Kromasil C_(18)(5μm,250mm×4.6mm)分析柱;流动相:甲醇-乙腈-0.003mol·L^(-1)磷酸二氢钾-二乙胺(55:25:20:0.06);流速1.0mL·min^(-1);检测波长为230nm。结果:精密度和稳定性均良好;粉防己碱浓度在1.392~11.14μg·mL^(-1)范围内与峰面积呈良好的线性关系,r=0.9999,平均回收率为98.4%;防己诺林碱浓度在0.575~6.90μg·mL^(-1)范围内与峰面积呈良好的线性关系,r=0.9998,平均回收率为97.9%。结论:本方法简便、专属,重复性好,可用于迪可颗粒中粉防己碱和防己诺林碱的含量测定。 Objective:To develop a method for determination of tetrandrine and fangchinoline in Dike granules. Methods: Using RP - HPLC method with Kromasil Cls (5 μm, 250 mm × 4. 6 mm) column, the mobile phase was a mixture of methanol - acetonitrile - 0. 003 mol · L^ -1 potassium dihydrogen phosphate - diethylamine ( 55 : 25 : 20: 0. 06) ,the flow rate was 1.0 mL · min^-1 ,with the detection wavelength at 230 nm. Results:The precision and stability were fine. The calibration curves were linear in the range of 1. 392 - 11.14 μg· mL^-1 for tetrandrine (r = 0. 9999, n = 5 ) and 0. 575 - 6. 90 μg · mL^- 1 for fangchinoline ( r = 0. 9998, n = 5 ). The average revovery of tetrandrine was 98.4% and that of fangchinoline was 97.9%. Conclusion :The method is convenient, selective and reproducible for determination of tetrandrine and fangchinoline in Dike granules.
