摘要
用荧光光谱法研究水溶液中氧氟沙星与小牛胸腺DNA的结合作用,测定了两个不同温度下氧氟沙星与DNA的结合常数KA和结合位点数n,探讨了其荧光猝灭机制;根据热力学参数确定了氧氟沙星与DNA之间的作用力以静电作用为主;进一步考察了二种金属离子及盐效应对该结合反应的影响.
The interaction of ofloxacin and calf thymus DNA has been studied by fluorescence spectroscopy (Tris-HCl buffer, pH = 7.0). The results show that the fluorescence intensity of ofloxacin at 485nm is quenched when DNA is added. The quenching mechanism of fluorescence of ofloxacin by DNA is a static quenching procedure, and the binding constant KA and the number of binding sites n are calculated. The main binding force between ofloxacin and DNA is electric charges interaction. The effect of two kinds of metal ion and salt effect on the interaction between ofloxacin and DNA is also studied.
