摘要
目的:探讨血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)体外对不育症精子运动参数和胞内钙离子的影响。方法:不同浓度的AngⅡ在体外与经上游优化处理的不育症精子分别共同孵育20、40min,采用计算机辅助精子分析(CASA)检测人精子运动参数;荧光分光光度计检测胞内游离Ca2+荧光强度的变化。结果:AngⅡ(10nmol/L和100nmol/L)在体外能显著提高不育症精子活率及运动参数(p<0·05),并激发胞内Ca2+快速升高(p<0·001);Losartan(AngⅡ受体AT1拮抗剂)、EGTA可以明显阻断和抑制这些过程。结论:AngⅡ在体外可以显著增高不育症患者精子的运动参数,其结果可能是通过人精子膜上的AT1受体结合作用后使胞外Ca2+内流而实现的。
Objective:To investigate the effects of angiotensin Ⅱ ( Ang Ⅱ ) on human sperm motility and intracellular calcium in vitro. Methods:Sperms were aseptically obtained by masturbation and prepared by swim -up technique from 32 infertile males, and then were incubated with different concentrations of Ang Ⅱ at 37℃ in vitro for 20 and 40 minutes respectively. Motility parameters were measured with computer - aided sperm analysis (CASA) and fluorescence intensity of intracellular Ca^2+ was measured by fluorospectrophotometer. Results:The results showed that Ang Ⅱ ( 10nmol/L or 100nmol/L) could significantly increase the motility parameters of male infertility spermatozoa incubated in vitro ( p 〈 0.05 ) and stimulate the increase of the intracellular Ca^2+ concentration( p 〈 0. 001 ). However ,these effects of Ang Ⅱ could be inhibited by Losartan(a Ang Ⅱ blocker) and EGTA. Conclusion:Ang Ⅱ (10nmol/L or 100nmol/L)can significantly increase the mobility of male infertility sperm in vitro. The mechanism may be explained by the influx of extracellular calcium caused by the integration of Ang Ⅱ with AT1 on human spermatozoa.
