摘要
目的探讨用血小板/T细胞活化抗原1(PTA1/Ig,亦简称CD226)融合蛋白预作用血小板后观察其对内皮细胞的黏附的阻断作用。方法妊娠高血压综合征患者血清与血管内皮细胞进行共培养,再与血小板进行粘附试验阻断实验,用间接免疫荧光法进行标记,用流式细胞仪检测受刺激后血管内皮细胞上血小板/T细胞活化抗原1的表达。结果妊娠高血压综合征患者和正常孕妇血清刺激的血管内皮细胞表达CD226的百分率分别为15.51%和7.15%,妊娠高血压综合征患者组显著高于对照组(P<0.001)。PTA1/Ig融合蛋白预作用于活化血小板后可明显地阻断其与内皮细胞的黏附,阻断率为48.9%,明显高于PTA1/Ig融合蛋白预作用于活化内皮细胞后对两者的阻断作用(阻断率为13.6%)。结论妊娠高血压综合征患者血清具有刺激血管内皮细胞表达CD226的作用,内皮细胞与血小板的黏附过程中以血小板表面的T细胞活化抗原1(PTA1)配体与内皮细胞表面的PTA1分子相互作用为主。
Objective To discuss adhesion blocking experiment of vascular endotelial cells with platelets. Methods The expression,regulation and adhesion of CD226 on vascular endothelial cells incubated by sera from 10 PIH patients and 10 normal pregnant women repectivdy was detected by flow cytometry, then incubated with inter- acted platelets by PTA1/1g protein. Results The positive rate of CD226 in PIH group was significandy higher than that in normal pregnant woman group(P 〈 0. 001 ). After the incubation with platelets, the positive rate of CD226 in PIH group was lower than before(P 〈 0.05 ). The blocking rate of interacted platelets by PTA1/1g protein to endotelial cells was 48.9 % , which was significantly higher than that of interacted endotelial cells by PTA1/1g protein to platelets. Conelusions The serum of PIH can improve the CD226 express of vascular endothelial cell. The PTA1 on the surface of endothelial cell combines with its legend on the platelets major in the adhesion.
