摘要
目的建立幼鼠结肠炎模型,研究其自然病程的特点,为临床研究儿童炎症性肠病(IBD)提供动物模型。方法采用三硝基苯磺酸(TNBS)/乙醇法制作幼鼠结肠炎模型,对照组给予生理盐水灌肠,0时组作为基础值不予任何刺激。应用统一评分标准对结肠粘膜损伤进行评估。结果模型组于造模后24h粘膜损伤最重,有明显溃疡形成,1周溃疡基本愈合,2周起粘膜大体评分与对照组差异无显著性,4周时镜下评分与对照组差异无显著性;模型组体重增长缓慢。结论幼鼠实验性结肠炎模型不同于成年鼠模型,可有生长发育迟缓的表现,更适合于对儿童IBD的研究。
Objective To establish a young rat model with experimental colitis and to explore the characteristics in injury and repair of the intestinal mucosa for the study of inflammatory bowel disease (IBD) in children via the animal model. Methods Afetr fasting 48 hours,a young rat model with colitis was established on anaesthesia with pentobarbital by injecting enterally 150 mg/kg of 2, 4, 6-trinitrobenzene sulfonic acid(TNBS) dissolved in 0.25 ml of 50% ethanol. Rats in control group were enterally given normal saline and rats in another control group were given nothing for the detection of basic standard. Injurious grade of intestinal mucosa was scored by Morries' and HS criteria. Results The obvious ulceration was observed in the intestinal mucosa at 24 hours after being given TNBS. The ulcer healed basically about a week later. No significant differences in injurious scores were observed two weeks later in the gross and four weeks later under microscope, respectively. The weight gained slowly in rats with colitis during experimental period. Conclusions It is feasible to study childhood IBD on the young rat model of colitis that is different from adult rat model of colitis due to concomitant growth retardation in young model.
