超声体外诱导白血病细胞K562凋亡的研究 被引量:1

摘要 目的研究超声对诱导白血病细胞K562凋亡的作用,为超声治疗的临床应用提供实验依据。方法以人慢性粒细胞白血病急变细胞株K562为研究对象,用频率为1.8MHz的超声波辐照白血病细胞K562,用光学显微镜观察凋亡细胞形态学改变,流式细胞仪检测细胞凋亡率。结果超声辐照K562细胞后24h,8mV×5min组及8mV×10min组细胞凋亡率分别为2.3%和8.28%;10mV×5min及10mV×10min组细胞凋亡率分别为9.36%和21.46%。电压为7mV时,超声辐照K562细胞后0.5h,1h,3h,5h细胞凋亡率分别为1.9%,3.6%,5.7%,7.7%。同时光学显微镜下可见凋亡小体形成等典型形态学改变。结论超声可以诱导白血病K562细胞凋亡。 Objective To observe the effect of ultrasound on apoptosis of K562 cells. Methods Human leukemia cell line(K562) cultured in vitro are treated by uhrasound at a frequency of 1.8 MHz. Apoptosis cells were evaluated by light microscope and detected using the flow cytometry assay (FCM). Results The percentage of apoptosis in cells exposure to 8 mV×5 min and 8 mV×10 min were 1.1% and 2.6 % respectively, 10 mV×5 min and 10 mV ×10 min were 9.7 % and 21.6 % respectively. The percentage of apoptosis in cells exposure to 7 mV×30 sec at 0.5 h, 1 h, 3 h, 5 h posttreatment were 1.9 %, 3.6 %, 5.7 %, 7.7 % respectively; Typical morphological changes of apoptosis such as apoptotic body were found under light microscope. Conclusion Ultrasound can induce apoptosis in vitro in human leukemia cell line K562.
