1株抗菌植物内生菌EJH-2菌株的分离和鉴定 被引量:31

摘要 目的从中药植物金银花的组织中分离到1株具有抗菌活性的植物内生菌EJH-2菌株,并对其进行了分子生物学鉴定。方法通过总DNA提取,PCR扩增,得到1 300 bp的16S rRNA序列。PCR产物序列通过BLAST软件在NCB I网站中进行同源性比较。通过B ioed it 7.0和Treedraw ing软件绘制系统发育树。结果EJH-2的16S rRNA序列和数据库中的类多粘芽胞杆菌KCTC 1663菌株的序列的同源性为99.76%。在系统发育树中,EJH-2菌株和多粘类芽胞杆菌在同一分支。结论EJH-2菌株应归属于多粘类芽胞杆菌(Paenibacillus polymyxa)。 Objective One endophyfic strain EJH-2 with strong antimicrobial activity was derived from tissue ot Lonicera japonica . Methods Thunb and identified based on 16S rRNA gene sequences. The sequence fragment of 1 300 bp 16S ribosomal RNA was amplified from total DNA of strain EJH-2 by PCR. The sequence analysis was down by the application of BLAST on the websites of NCBI and the phylogenctic tree was drawn by Bioedit 7.0 and Treedrawing. Results EJH-2 shared 99.76% 16S rRNA sequence homology with Paenibacillus polymyxa KCTC 1663. EJH-2 and Paenibacillus polymyxa were in same branch of phylogenetic trees. Conclusion The strain EJH-2 belongs to Paenibacillus polymyxa.
