鸡病毒性关节炎免疫扩散试验研究

Studies on the Immuno-diffusion Test for Detection of Viral Arthritis in Chickens

利用Vero细胞、成纤维细胞制备琼扩抗原，系统规定了鸡病毒性关节炎琼扩试验所需的最佳缓冲液，琼脂糖浓度、孔径、孔距、抗原保存条件。结果表明以pH8.1、0．06M巴比妥钠-HCl缓冲液，1％琼脂糖，孔径和孔距均为3mm，效果最佳。抗原少量分装-20℃保存，避免反复冻融。特异性试验表明，与其他相关病阳性血清均不产生交叉反应。

AGID antigens prepared with vero cells and Fibroblast were used to determine the optimum buffer system,gel concentration, well diameter and distance between wells in AGID for Viral Arthritis and conditions for storing antigens. It was shown that 3 mm of well diameter and distance and 1 % agarose yielded best results. Antigen should be stored in aliquots at -20℃ and avoid repeated freezing and thawing. Specificity test proved that it produced no cross reaction with the sera to other related diseases.
