摘要
目的观察血管紧张素Ⅱ受体2型(AT2-R)基因+1675A/G多态性及醛固酮合酶(CYP11B2)基因-344C/T多态性与高血压左室肥厚(LVH)的相关性。方法按心脏超声特点将314例原发性高血压患者分为左室肥厚〔LVH(+)〕组156例(男61例,女95例),无左室肥厚〔LVH(-)〕组158例(男96例,女62例)。使用常规方法提取白细胞DNA,采用多聚酶链式反应(PCR),限制性内切酶方法测定AT2-R基因多态性及CYP11B2基因多态性。结果(1)在LVH(+)组与LVH(-)组,男性和女性AT2-R基因+1675A/G多态性各基因型构成及等位基因频率间差别均无显著性意义(P>0.05);(2)CYP11B2基因-344C/T多态性显示LVH(+)组TT基因型频率显著高于LVH(-)组(P<0.05),而CT基因型频率LVH(+)组显著低于LVH(-)组(P<0.05);T等位基因频率LVH(+)组显著高于LVH(-)组(P<0.05)。结论(1)AT2-R基因+1675A/G多态性与高血压LVH的发生无关;(2)CYP11B2基因-344TT基因型及T等位基因频率与高血压LVH的发生有关。
Objective To evaluate the relationship between gene polymorphism of angiotensin Ⅱ type 2 - receptor ( AT2 - R), aldosterone synthase (CYP11B2) and left ventricular hypertrophy (LVH) in hypertension (EH). Methods 314 patients with EH were studies, whom were divided into two groups on base echocardiography characteristic: the left ventricular hypertrophy [LVH ( + ) ] group was 156 patients (man 61 cases, women 95 cases) , and the no left ventricular hypertrophy LVH ( - ) ] group was 158 patients (man 96 cases, women 62 cases). DNA was extracted from white blood cell. polymerase chain reaction (PCR) and Hae Ⅲ restriction endonuclease digestion were employed to study + 1675G/A polymorphism of AT2 - R gene and - 344C/T polymorphism of CYP11 B2 gene. Results ( 1 ) There was no significant difference in + 1675 G/A polymorphism of AT2 - R gene between the two groups. ( 2 ) The frequency of the - 344TT genotype, T allele in LVH ( + ) group was significantly higher than that in the LVH ( - ) group ( P 〈 0. 05 ) ; and the frequency of the - 344CT genotype in LVH ( + ) group was significantly lower than that in the LVH ( - ) group ( P 〈 0. 05 ). Conclusion ( 1 ) There was no significant relationship between + 1675G/A polymorphism of AT2 - R gene and the occurrence of LVH. (2) A significant relationship between the frequency of -344TT genotype, T allele and LVH in EH could be found.
