摘要
目的探讨实验性脑梗死大鼠脑组织单核细胞趋化蛋白-1(MCP-1)的表达及其意义。方法将健康雄性SD大鼠42只随机分为假手术组和脑梗死组。参照longa等方法制成大鼠局灶性大脑中动脉阻断(MCAO)模型,于术后6 h、24 h、48 h、72 h、7 d分别处死动物,检测脑组织的含水量,病理切片HE染色观察组织病理学改变,免疫组织化学的方法测定MCP-1的阳性细胞数。结果脑梗死后脑组织MCP-1表达于6 h开始增多,24 h达到高峰,48 h仍高于假手术组,7 d明显减少,与梗死灶周围单核/小胶质细胞浸润及脑水肿变化规律基本一致。结论脑梗死后脑内MCP-1的表达与脑水肿形成有关,且促进了单核巨噬细胞的聚集和小胶质细胞的活化,其参与了脑缺血损伤后的炎症免疫反应。
Objective To investigate the expression and role of monoeyte ehemoattraetant protein - 1 ( MCP - 1 ) in the brain of middle cerebral artery occlusion (MCAO) rats. Methods Forty - two healthy male Sprague - Dawley rats were randomly divided into sham operation group and cerebral infaretion group, The model with thread embolism of MCAO in rats was made. At the time of 6 h, 24 h, 48 h, 72 h and 7 d after MCAO, the changes of brain water content were measured; the changes of histopathology were observed by the HE staining method; immunohistoehemistry method was adopted to detect the changes of MCP - 1 expression. Results In the isehemie brain, the expression of MCP - 1 increased at 6 h and reached maximum at 24 h, and deereased obviously at 7 d. The regularity of monoeyte/mieroglia infiltration and brain edema was identieal with the one mentioned above, Conclusion The expression of MCP - 1 may play an important role in brain edema formation, and it might promote the collection of monoeyte and the activation of mieroglia, participate in the inflammation and immune process in the isehemie brain.
