健心方防治心力衰竭分子机理研究

摘要 [目的]探讨了健心方药物血清对于心肌细胞NF-kB活性的影响,阐明健心方保护心肌的作用机理。[方法]采用荧光素酶报告基因方法检测H2O2处理心肌细胞NF-kB活性的改变及丹参酮对损伤后心肌细胞NF-kB活性的影响。[结果]荧光素酶报告基因检测结果表明H2O2可明显增强心肌细胞NF-kB活性(P<0·05),而健心方药物血清可抑制这种激活作用(P<0·05);大小剂量的药物血清均可拮抗H2O2对心肌细胞的损伤作用(P>0·05)。[结论]心肌细胞损伤后NF-kB的激活是CHF发生发展中的重要过程,健心方可拮抗这种激活作用;健心方的心肌保护作用是通过对异常增高的NF-kB活性的拮抗产生的。 [Objective] Explore the effect of the medicinal serum of jianxin Fang on cardiac muscle cell NF-κB activity, expound the functional mechanism of Jianxin Fang in protecting cardiac muscle. [Method]Use fluorescein enzyme to report gene, measure the activity of NF-κB dealt with H2O2 and the influence of tanshinone on injured NF-κB. [Result] The fluoreseein enzyme report shows that H2O2 can obviously increase NF-κB activity, and medicinal serum of Jianxin Fang can inhibit this activation; large or small dosage of medicinal serum can resist the injury of H2O2 on cardiac muscle cell. [Conclusion] After injury of cardiac muscle cell, activation of NFkB is the key course of CHF development, which can be inhibited by Jianxin Fang; the protection of Jianxin Fang on cardiac muscle cell is made through resisting the much increased activity of NF-κB.
