电针对抑郁模型大鼠糖水摄入量和性行为的影响及机理研究 被引量:9

摘要 目的:探讨电针对抑郁状态下奖赏效应的调节作用和作用机理。方法:选用SD雄性大鼠,分为对照组、模型组、电针组。用慢性应激结合孤养的方式造模。电针组取“百会”“印堂”穴,使用HANS LH202型电针仪进行电针。慢性应激程序开始前和结束后分别测量大鼠蔗糖水摄入量;造模、治疗周期结束后计算大鼠15 min舔吻次数和跨骑次数;采用高效液相-电化学法测定部分脑区多巴胺(DA)含量。结果:电针组大鼠糖水摄入量明显多于模型组(P<0.05);电针组大鼠舔吻次数和跨骑次数与模型组相比显著增加(P<0.05);电针组额叶皮层、纹状体、下丘脑DA含量显著升高,额叶皮层、下丘脑内DA含量与模型组比较有显著性差异(P<0.05);纹状体内DA含量与模型组比较有极显著性差异(P<0.01)。结论:电针可以影响慢性应激抑郁模型大鼠食物奖赏效应和性奖赏效应,并可能通过调节中枢不同脑区DA参与抑郁状态下奖赏效应的恢复。 Objective: To investigate the effect of electroacupuncture (EA) on the natural reward behavior and the related mechanism in depression rats. Methods: Thirty male SD rats were evenly randomized into control, model and EA groups. Depression model was established by using chronic stress stimulation including forced cool water swim, tail-clamping, fasting, electric shock, etc., once daily, 21 days altogether. One hour before stress stimulation in EA group, EA (2 Hz, 2 mA) was applied to "Baihui" (GV 20) and "Yintang" (EX-HN 3) for 20 min, once daily, continuously for 21 days. The consumption of the rats' sucrose intake was detected before and after establishment of depression model. The rat's sexual behavior was recorded by calculating the times of mounting the female rat and sniffing female rat's pudendum in 15 minutes. Dopamine (DA) contents of frontal cortex, striate body and hypothalamus tissues were assayed by using high performance liquid chromatography electro-chemical detection technique. Results: Compared with control group, the male rats' sucrose intake amount, pudendum-sniffing times, DA contents of frontal cortex, striate body and hypothalamus tissues in model group decreased significantly (P〈0.05, 0.01) ; while compared with model group, the rats' sucrose intake amount, mounting and sniffing times and DA contents of the aforementioned 3 cerebral areas in EA group increased significantly ( P 〈 0.05). No significant differences were found between control and EA groups in sucrose intake amount, pudendum-sniffing times and DA contents of the 3 cerebral areas (P〉0.05). Conclusion: EA can promote recovery of the rats' food rewarding and sexual reward and adjust the secretion of DA in depression rats.
