摘要
目的观察幽门螺杆菌(H.pylori)作用于胃癌细胞后线粒体编码基因COXⅠmRNA及蛋白的表达变化,探讨H.pylori致胃癌的分子机制。方法H.pylori培养滤液分别作用于胃癌细胞4、8和12 h后,收集细胞,应用RT-PCR和W estern b lot法检测各时相点COXⅠ基因和蛋白表达变化。结果H.pylori培养滤液作用4 h后,线粒体COXⅠmRNA的表达开始降低,8 h下降更明显,12 h下降速度减缓。各时相点的表达量明显低于对照组(P<0.05)。COXⅠ蛋白表达呈现出相同的趋势,各时相点的表达量明显低于对照组(P<0.05)。结论H.pylori可下调胃癌细胞线粒体编码基因COXⅠmRNA及蛋白的表达,影响线粒体功能。
Objective To study the effects of concentrated H. pylori culture supematant (CHCS) on the expressions of COXⅠ mRNA and protein in gastric carcinoma cells, and explore the potential role of H. pylori in the gastric carcinogenesis. Methods After SGC7901 ceils were treated by 6 μl/ml CHCS for 4 h, 8 h, 12 h respectively, the expression of COXⅠmRNA and protein was detected by RT-PCR and Western blotting. Results The expression of COXⅠ mRNA decreased gradually after exposure to CHCS for 4 h. Especially after 8 h, the expression decreased significantly. When treated with CHCS for 12 h,the expression continued to decrease (P 〈 0.05 ). The expression of COX Ⅰ protein revealed the same trend. Conclusion H. pylori could affect mitochondrial function by down-regulating the expression of mitochondrial encoding cytochrome oxidase Ⅰgene.
