脂溢性角化病皮损的表皮中TGF-βⅠ型和Ⅱ型受体表达下调 被引量:9

摘要 目的探讨转化生长因子-β受体(transform ing growth factorβreceptor,TGF-βR)在脂溢性角化病皮损中表达水平的变化及其意义。方法采用实时定量聚合酶链式反应(quantitative real-tim e PCR)和S-P免疫组织化学技术分别检测脂溢性角化病及正常对照皮肤中I型和Ⅱ型TGF-β受体(TGF-βRⅠ,TGF-βRⅡ)的表达。结果TGF-βRⅠ和TGF-βRⅡ在脂溢性角化病皮损表皮中的表达较正常表皮显著降低。结论脂溢性角化病皮损中表皮TGF-β受体表达下调有助于表皮细胞的异常增生,促进了该表皮肿瘤的形成。 Objective To investigate the expression and significance of transforming growth factor beta receptors (TGF-βRⅠ ,TGF-βRⅡ ) in the epidermis of seborrheic keratosis (SK). Methods Twenty-five pieces of SK skin including 14 from face and 11 from trunk were from 25 SK patients, 11 male and 14 female, mean age 57.3 years, whose average course of disease was 3.7 years. Another 24 of matched location were from 24 healthy individuals underwent plastic surgery, 10 male and 14 female, average age 44 years. All pieces of skin were divided into two parts, one for SP immunohistochemisty and one for real-time PCR. Quantitative real-time PCR and SP immunohistochemical techniques were utilized to assess the expression of TGF-βR Ⅰ and TGF-βR Ⅱin the epidermis of seborrheic keratosis and normal control skin. Results The two methods respectively proved the mRNA and protein expression levels of TGF-βR Ⅰ and TGF-βR Ⅱin the epidermis of seborrheic keratosis were significantly reduced in comparison with those of normal control skins. Conclusion Downregulation of TGF-β receptors may interfere with TGF-β signaling, and contribute to formation of aberrant hyperplastic epidermis in seborrheic keratosis.
