神经干细胞分化过程中微管蛋白表达变化的光电镜观察 被引量:3

摘要 目的对神经干细胞向神经元定向分化过程中微管蛋白的表达变化进行光、电镜观察研究。方法采用细胞培养技术、免疫荧光技术以及免疫电镜技术对神经干细胞向神经元定向分化过程中微管蛋白的表达变化进行观察。结果在神经干细胞向神经元定向分化的不同时期,存在微管蛋白的表达变化,在分化初期以核周附近分布明显,随神经元的成熟散在分布于胞质中及突起内,形成细网状,构成细胞骨架,维持细胞形态。结论在神经干细胞向神经元定向分化过程中伴随有微管蛋白的表达变化,随神经元的成熟而构成细胞骨架,维持细胞形态。 Objective To observe and study the expression changes of β-tubulin in neuron-oriented differentiation process of neural stem cells with optical and electron microscopes. Methods Using the techniques of cell culture, fluorescent immunocytochemistry and immunoelectron microscopy, we observed the expression changes of tubulin in neuron-oriented differentiation process of neural stem cells. Results During the different phases of the differentiation process there were expression changes of tubulin: in the early phase it showed an obvious distribution around nuclei, and then, with the maturation of neurons, it was scattered in cytoplasm and processes to form thin network, composing the cell skeleton and sustain the cell shape. Conclusion Expression changes of tubulin are concomitant with the neuron-oriented differentiation process of neural stem cells, and, with the maturation of neurons, it composes the cell skeleton and sustain the cell shape.
