转化生长因子β_1对培养的瘢痕成纤维细胞增殖的调控作用 被引量:7

摘要 目的探讨TGF-β1对瘢痕成纤维细胞增殖的调控作用。方法采用细胞培养、MTT法、流式细胞仪和免疫组化方法观察TGF-β1在不同作用剂量和时间对增生性瘢痕来源的成纤维细胞的增殖影响。结果在20%胎牛血清条件下TGF-β1可刺激瘢痕成纤维细胞增殖,其作用在10ng-50ng/ml之间呈剂量效应关系;50ng/ml TGF-β1作用12h开始增殖显著,并可持续到72h。细胞周期分析表明:在TGF-β1作用下,G0/G1%呈降低趋势,而S%呈升高趋势,反映细胞增殖活力的PI值也呈升高趋势。免疫组化结果也显示增殖细胞核内抗原(PCNA)随用药浓度增加而逐渐增加。说明TGF-β1对体外培养的瘢痕成纤维细胞具有促增殖作用。结论TGF-β1可能通过调控成纤维细胞的增殖而在增生性瘢痕的形成过程中起重要作用。 To investigate the effects of transforming growth factor-β1 (TGF-β1) on growth and the proliferation of cultured human fibroblasts isolated from hypertrophic scars. Methods The effect of TGF-β1 at different dosage and time was observed by using MTT, its effect on cell cycle and proliferation index (PI) (SA-G2M) was checked by flow cytometry (FCM), and its effect on proliferation cell nuclear antigen (PCNA) was assessed by immunohistocytochemistry. Results With 20% fetus cow serum, TGF-131 stimulated cell proliferation, with a dose-dependent increase at concentrations 10-50ng/ml. At 50mg/ml, significant cell proliferation began from 12h and lasted until 72h. Cell cycle assessment indicated lowering inclination of G0/ G1% and elevating inclination of S%. Proliferation index (PI) and PCNA , which suggest cell proliferation activity, were elevated especially when the concentration was greater than 10ng/ml. Conclusion TGF-β1 may exert its function on the formation of hypertrophic scars by stimulating fibroblast proliferation.
