脊柱黄韧带骨化细胞COX2、VEGF的表达及其意义 被引量:6

EXPRESSION AND CHINICAL SIGNIFICANCE OF COX2 AND VEGF IN CELLS OF SPINAL LIGAMENTUM FLAVUM OSSIFICATION

摘要 目的探讨脊柱黄韧带骨化韧带细胞COX2、VEGF表达及其临床意义。方法8例胸椎黄韧带骨化症手术取得的黄韧带标本,做COX2、VEGF免疫组化染色,分析两者表达及相互关系;原代培养黄韧带细胞,分常规(N)、低氧(O)、塞来昔布药物作用(D)三组,做COX2、VEGF免疫细胞化学染色和ELISA测定上清液VEGF。结果8例中,6例共阳性表达COX2和VEGF,2例COX2阳性和VEGF弱阳性,直线相关分析吸光度值,相关系数r=0.8461(P<0.05)。N组细胞COX2阳性表达,VEGF弱阳性,O组两者均强阳性,D组10μmol/L两者弱阳性,20-100μmol/L均阴性。N组上清液VEGF浓度(5.379±0.245)mg/L,显著低于O组的(13.256±0.138)mg/L,D组20-100μmol/L各组浓度显著低于对照组。结论脊柱黄韧带骨化细胞异常分泌COX2和VEGF,促进局部血管新生,是骨化必备条件。选择性COX2抑制剂可能控制疾病发展或防止术后复发。 Objective To discuss the expression and clinical significance of COX2 and VEGF in cells of spinal ligamentum flavum ossification. Methods 8 samples of thoracic ossification of ligamentum flavum (TOLF) were obtained during operation. COX2 and VEGF immunohistochemistry (IHC) was performed, and the expression and relationships were analyzed. Ligamentum flavum cells (LFCs) were cultured under normal (N), hypoxia (O) and celebrex-treated (D) condition respectively, and then COX2, VEGF immunocytochemieal staining and ELISA test of supernatant VEGF were applied. Results Six of eight eases expressed both COX2 and VEGF positively, while 2 eases expressed COX2 positively and VEGF weakly. The absorbency (A) value was analyzed, and the correlation coefficient r was 0. 8461 (p〈0. 05). LFCs expressed COX2 positively and VEGF weakly in group N and expressed both most intensely in group 0. LFCs expressed both weakly under 10μmol/L and negatively under 20-100μmol/L in group D. The concentration of VEGF in group N was (5.379±0.245) mg/L, lower than that in group O (13.256± 0. 138) mg/L. The concentration in group D treated with 20-100μmol/L was lower than that in the control group. Conclusion LFCs from TOLF secrete COX2 and VEGF abnormally and stimulate vasculogenesis, which is indispensable to ossification. The selective COX2 inhibitor may prevent the aggravation and recurrence after operation.
