卵巢癌细胞与腹膜间皮细胞相互作用对腹膜粘附和侵袭的影响

摘要 目的:探讨卵巢癌细胞与腹膜间皮细胞(HPMC)相互作用对腹膜粘附和侵袭的影响。方法:用卵巢癌细胞SKOV3条件培养液(SKOV3-CM)或SKOV3-CM加转化生长因子β1(TGF-β1)中和抗体培育HPMC12h,用粘附实验和Boyden小室侵袭实验检测癌细胞对HPMC的粘附和侵袭,并采用ELISA检测各组HPMC上清中纤维粘连蛋白(FN)及透明质酸(HA)蛋白水平。结果:SKOV3-CM培育HPMC后,癌细胞的粘附率为(48.7±3.7)%,侵袭的细胞数为118.2±10.7,HPMCFN及HA蛋白分别为(475.69±9.54)ng/ml,(368.92±16.15)ng/ml,均高于空白对照组(P<0.01)。SKOV3-CM中加入TGF-β1中和抗体培育HPMC后,癌细胞的粘附率为(36.3±2.8)%,侵袭的细胞数为73.4±6.5,HPMCFN及HA蛋白分别为(287.17±10.07)ng/ml,(219.64±12.56)ng/ml,与SKOV3-CM组相比,均明显降低(P<0.01)。结论:卵巢癌细胞分泌TGF-β1,可以促进癌细胞与HPMC的粘附,有利于癌细胞的腹膜转移,促进HPMCFN及HA的表达可能是其作用的分子机制之一。 Objective: To study the impact of interactions of ovarian carcinoma cells and human peritoneal mesothelial cells(HPMCs) on adhesion and invasion of ovarian carcinoma to peritoneum. Methods: HPMCs were pretreated with ovarian carcinoma cell SKOV3 conditioned medium (SKOV3-CM) for 12 hours in the presence or absence of transforming growth factorβ1 (TGF-β1) neutralizing antibody. Adhesion and invision of carcinoma cells to HPMCs were detected by adhension assay and Boyden Chamber invasion assay. The expressions of fibronectin(FN) and hyaluronan(HA) were studied by ELISA. Results: After HPMCs being preincubated with SKOV3-CM, adhesion rate of carcinoma cells was (48.7±3.7)% and the number of invasive carcinoma cells was 118.2±10.7. FN and HA protein expressions of HPMCs were (475.69±9.54) ng/ml and (368.92±16.15) ng/ml, which were increased compared with those in the control group (P〈0.01). After HPMCs being preincubated with SKOV3-CM and TGF-β1 neutralizing antibody, adhesion rate of carcinoma cells was (36.3±2.8)% and the number of invasive cartinoma cells was 73.4±6.5, FN and HA protein expressions of HPMCs were (287.17±10.07) ng/ml and (219.64±12.56) ng/ml, which were inhibited compared with those in SKOV3-CM group (P〈0.01). Conclusions: Ovarian carcinoma cells can promote tumor invasion by enhancing carcinoma cell-HPMC adhesion through TGF-β1, and one of its action mechanisms is to promote the expression of FN and HA of HPMC.
