苯丁酸钠对喉癌Hep-2细胞株的体外诱导分化作用 被引量:1

摘要 目的:探讨诱导分化剂苯丁酸钠(sodiumphenylbutyrate,PB)在体外对喉癌Hep-2细胞株的生长和分化的影响。方法:应用细胞生长曲线测定法、软琼脂克隆形成试验、细胞形态观察、划痕试验、MTT法、流式细胞术等对在体外经不同浓度PB处理过的Hep-2细胞进行检测,观察细胞分化程度及运动能力的改变。结果:PB在一定浓度范围内能明显抑制Hep-2细胞的生长,作用3d的半数抑制浓度(IC50)为5.32mmol/L,与剂量和时间呈依赖性关系;PB能诱导Hep-2细胞分化,使其克隆形成能力减弱,细胞运动能力下降,细胞突起增长,G1→S期阻滞。结论:PB可以诱导喉癌Hep-2细胞分化,并使细胞的运动能力下降,为其分化治疗喉癌提供了实验依据。 Objective: To study the inducing effect of sodium phenylbutyrate (PB) on laryngeal carcinoma cells Hep-2 in vitro. Methods: Soft agar clone forming test, cell morphology, scratch test, MTT and flow cytometry were used to examine the changes of Hep-2 cells treated by different concentration of PB in vitro. Results:There was a dose-dependent and time-dependent inhibition of the cell proliferation by PB. The IC50 after 3 days was about 5.32 mmol/L. Hep-2 cells exposed tension of cell processes, and increasing of GI fraction with a inhibition.
